Anti-ulcer effect of cinnamon and chamomile aqueous extracts in rat models

Aim of the work: To evaluate antiulcer potency of aqueous extracts of cinnamon (CINA) or chamomile (CHAMO) (100, 200, 300 or 400 mg/kg of body weight) and to compare the antiulcer potency with equivalent potency obtained with the pharmaceutical Zantac ™ (Ranitidine) drug

Design of the study: The rat feeding study consisted of 50 male albino rats weighing 160±5g . Five rats served as positive control ; another group of five rats served as standard group. The remaining 40 rats were divided into two groups of equal number. The first group was treated with aqueous extracts of CINA; whereas the second group was treated with aqueous extract of CHAMO. The CINA group was further divided into four subgroups given oral dosages of 100, 200, 300 or 400 mg /kg of body weight. The CHAMO group was divided into four subgroups treated in an identical manner as described above . The oral dosages were given daily via tube feeding for seven consecutive days. At the last day of the trial, the rats were fasted for 24 hours with access of water. The standard group was given Zantac (ranitidine) intraperioteneally (IP) at a dose of 6 mg/100 g body weight, 60 min prior to the administration of ethanol. Rats were given oral saline and aqueous extracts of cinnamon or chamomile 120 min prior administered ethanol. Ethanol was administered orally to all groups at 0.5 ml / 100 g body weight. After 4 hours, all rats were sacrificed by an overdose of diethyl ether and their stomachs removed and washed with saline. The pH value, the volume of gastric juice, the ulcer area, and curative ratio were estimated according to standard techniques. The stomach was separated, preserved in formalin solution (10 %) for subsequent mounting on paraffin film and preparation of sections.

Results: The pH value of the rats with induced stomach ulcer averaged 4.70±0.26. The treatment with Zantac ; CINA or CHAMO was associated with significant increase in the pH values compared to the respective mean value among the positive group. The mean volume of gastric juice among the standard (4.30±0.37ml) and the positive group (4.60±0.52ml) was almost comparable. Oral administration of CINA or CHAMO was associated with significant reduction in the mean volume of gastric juice compared to the respective normal value. Curative ratios of gastric ulcer were better in rats given CINA or CHAMO over those given Zantac. Furthermore, CHAMO was superior over CINA in its curative ratios of gastric ulcer. Histological examination showed marked necrosis of gastric mucosa associated with congestion of submucosal blood vessels, submucosal edema and hemorrhage in the stomachs of rats receiving ethanol. The stomachs of the group receiving Zantac showed necrosis of gastric mucosa associated with hemorrhage. Stomachs of rats treated with CINA at a dose of 100 mg or 200 mg showed atrophy of gastric mucosa associated with submucosal edema . Whereas, higher CINA dosages of 300 and 400 mg were adequate and histopathological changes disappeared from the stomachs . In the group receiving 100 mg CHAMO , there appeared submucosal leucocytic cells infiltration in the stomachs. The groups receiving increasing CHAMO dosages i.e., 200 ,300 or 400 mg showed no histopathological abnormailities.

Conclusion: Water extracts of cinnamon and chamomile had potential antiulcer effect, which was superior to the respective effect observed with Zantac. Chamomile extracts were more superior to cinnamon in its chemoprotective effect on the stomach lining . The antiulcer curative ratios were dose dependent with no adverse effects.
